How do you make create a heat-fixed bacterial smear?
Definition
1. With inoculating loop, place a small drop of water on a clean glass slide. 2. With sterilized (flamed) loop, remove a small amount of the bacterial colony from the culture and mix thoroughly with the water on the slide. 3. Spread the mixture creating two regions: a dense cellular populated region and a less dense region (as seen below). [image]
4. Let the slide air dry at room temperature. DO NOT HEAT. 5. Fix the air dried smear by quickly passing it three times through a flame until the slide is very warm to the touch.
