Indications: Psoriasis Arthritis, rheumatoid MOA:immunosuppressant: inhibits dihydrofolate reductase; inhibits lymphocyte proliferation (folate antagonist) - Methotrexate is an antimetabolite of the folic acid analog type. Methotrexate is cell cycle–specific for the S phase of cell division. Activity is due to inhibition of DNA, RNA, thymidylate, and protein synthesis as a result of relatively irreversible binding with dihydrofolate reductase, which prevents reduction of dihydrofolate to the active tetrahydrofolate. Growth of rapidly proliferating cells (epithelial cells in psoriasis, bone marrow, fetal cells, buccal and intestinal mucosa, cells of the urinary bladder, spermatogonia) is affected more than growth of most normal tissues and skin. |
