Term
Numerical Aperture Formula
Definition

NA = nsinθ

remember to half the aperature angle

Term
Limit of Resolution
Definition
The minimum distance or angular separation between two point objects which allows them to be resolved
Term
Limit of Resolution Formula
Definition
LR = (0.61 * λ)/(NA)
Term
Two things affecting limit of resultion
Definition
Wavelength and numerical aperature.
Term
What emits electrons into the vacuum in an electron microscope?
Definition
e-gun or filament
Term
How does the e-gun emit electrons into the vacuum?
Definition
The current heats the filament and electrons boil into the vacuum.
Term
What is the purpose of placing tissue in 30% sucrose?
Definition
Cryoprotection
Term
Refraction definition and cause
Definition

Light bending as it changes speed when it passes through the interface of two mediums.

Ex. glass/air or glass/oil

Term
Astigmatism
Definition
Where rays that propagate in two perpendicular planes have different foci. The vertical and horizontal lines are in sharp focus at two different distances.
Term
Constructive Interference
Definition
When the crest of one wave passes through, or is superpositioned upon, the crest of another wave, we say that the waves constructively interfere. Constructive interference also occurs when the trough of one wave is superpositioned upon the trough of another wave. Amplitudes increase.
Term
Destructive Interference
Definition
When the crest of one wave passes through, or is superpositioned upon, the trough of another wave, we say that the waves destructively interfere. Amplitudes subtract.
Term
Oil Immersion
Definition
A transparent oil of high refractive index, thereby increasing the numerical aperture of the objective lens.
Term
Fixation
Definition
Fixation preserves a sample of biological material (tissue or cells) as close to its natural state as possible in the process of preparing tissue for examination.
Term
Osmium tetroxide
Definition
Oxidation of macromolecules such as lipids leads to  atoms becoming cross-linked. It is also used by researchers to stain biological things in order to image them using transmission electron microscopy, by embedding a heavy metal directly into the cell.
Term
Glutaraldehyde
Definition
Glutaraldehyde is used in biological electron microscopy as a fixative. It kills cells quickly by crosslinking their proteins and is usually employed alone or mixed with formaldehyde[3] as the first of two fixative processes to stabilize specimens such as bacteria, plant material, and human cells. A second fixative procedure uses osmium tetroxide to crosslink and stabilize cell and organelle membrane lipids.
Term
Why you dont use Glutaraldehyde with fluorescent:
Definition
Glutaraldehyde can produce fluroscent properties and fuck your shit up.
Term
Which microscope requires a secondary fixative?
Definition
Electron microscope. Can be osmium tetroxide.
