That's what methotrexate, the folic acid analog, does. 5-Fluorouracil prevents the conversion of dUMP to dTMP by forming F-dUMP, binding 5,10-methylene THF/thamidylate synthase, and permanently inhibiting thamidylate synthase. Both methotrexate and 5-Fluorouracil should be administered at just sub-lethal doses.

It's involved in purine degradation and salvaging. Degradation: AMP à Adenosine à Inosine à Hypoxanthine (-R1P) à Xanthine (extra -OH by Xanthine Oxidase, generation of superoxide) à Uric Acid (again: extra -OH by way of Xanthing oxidase, generation of superoxide)

What's gout?
Definition
Elevated uric acid levels because purines are being over-produced. Can be caused by PRPP synthetase not responding to feedback inhibition by purine nucleoside diphosphatases or by deficiency of HGPRT leading to increased PRPP activity. Rx = Allopurinol (blocks production of uric acid).

What's Lesch-Nyhan Syndrom?
Definition
Complete HGPRT activity deficiency so hyoxanthine and guanine are not salvaged. Rx = allopurinol to stop uric acid production; however this doesn't help neurological symptoms, which remain largely unexplained. X-linked.

ADA = Adenosine Deaminase. Conversion to deoxyinositine and inositine isn't happening. White blood cells can't proliferate. Associated with Severe Combined Immunodeficiency (SCID). Tx = bone marrow transplant.
